INTRODUCTION TO PACKED CELL VOLUME (PCV)
Packed cell volume is a reasonable index of red blood cells population. PCV in Greek word 'Hematocrit' means Separation I.e to separate the components of blood.
PCV is the measure of relative mass of RBC presents in a sample of whole blood which is expressed as %
PCV is used to investigate when it is not possible to measure hemoglobin and diagnose Polycytemia (Having High Concentration of RBC) (Erythrocytosis)
PCV is the proportion occupied by Red blood cells in relation to the whole blood placed inside the capillary tube and centrifuged at a specified time.
Principle of PACKED CELL VOLUME
PCV is a % of a known volume of occupied by a packed redcells. When blood in the tube is centrifuged at a constant speed (3000 revolution) and period of time.
MATERIALS NEEDED FOR PCV
-Hematocrit
-Hematocrit Reader
-Bunsen flame
-Capillary tube
-Dry cottonwool
-EDTA Blood sample
FACTORS AFFECTING PCV RESULT
-Quality of capillary tube
-Speed and Time of concentration
-Improper sealing of capillary tube
-Clotted sample
-Anaemic patient
-Improper collection of sample
PROCEDURE FOR PCV
1. 3/4 of well anticoagulated blood was picked with a capillary Tube
2. The back of the capillary tube was wiped with dry cottonwool
3. The unfilled part of the capillary was sealed with a sealer (Or Bunsen burner or plasticin)
4. The capillary tube was placed in micro hematocrit
5. It was spinned at about 12,000 for 5 minutes
6. The result was read with micro Hematocrit reader and recorded in %
