LOCI - Laboratory of Cognitive Interventions

LOCI uses mobile technology to examine the automatic & subconscious reactions associated with drug use in real-time. The Laboratory of Cognitive Interventions (LOCI) examines the cognitive processes or biases associated with drug use and relapse.

Once these cognitive biases are identified, cognitive retraining interventions are developed to target (reduce) maladaptive cognitive biases. Cognitive retraining interventions, such as attentional retraining, can be delivered in the laboratory or using mobile devices such as personal digital assistants (PDAs) or Smartphones. Currently the laboratory is examining the efficacy of an attentional retraining intervention for smoking cessation delivered on a Smartphone. The laboratory is also investigating the utility of mindfulness-based interventions and pharmacological interventions that improve cognition. Ultimately, cognitive interventions might be delivered on a Smartphone when the individual is most in need of the intervention (EMI, Ecological Momentary Intervention).

WHY?
A vaccine can prevent reward feelings or craving relief from taking the target drug, and so reduces motivations and relapse.

WHAT?
What's challenging about making a anti-drug vaccine? The cells that make antibodies (B-lymphocytes) normally do not detect or respond to drugs.

HOW?
To solve the problem, a molecute hapten is made from the target drug to do 2 things: pull B-lymphocytes close, and teach it how to make antibodies!

1. "Pulls it close"
Hapten has a carrier protein that attracts B-lympohocytes. The high-affinity binding stimulates it to make antibodies.

2. "Teach it"
Hapten carries a "how-to" template for B-lymphocytes to make antibodies. After they bind ... BAM! The B-lymphocyte absorbs the template, turns into a self-cloning plasmocyte, which makes & releases lots of antibodies!
